Using ReMOT technology to genetically modify Culex tarsalis mosquitoes

The main objective of this project is to knock-out the Kynurenine monooxygenase (kmo) gene responsible for the eye color in Culex tarsalis by using ReMOT technology. In-vitro cleavage assay confirmed sgRNAs raised against exon 3 and 5 will be injected into 24h post blood fed females along with P2C-Cas9 and saponin. The offspring of the injected females will later be screened for any change in the eye color.
